ANCA-3-Line

The ANCA-3-Line immunoblot test is a membrane-based enzyme immunoassay for the semi-quantitative determination of antineutrophil cytoplasmic IgG antibodies (ANCA) against proteinase 3 (PR3), myeloperoxidase (MPO), and glomerular basement membrane (GBM) in human serum or plasma. It is used for the diagnosis of systemic autoimmune vasculitis.
ANCA associated vasculitis may be a fast progressing life threatening disease, thus rapid and reliable ANCA serology is particularly vital in vasculitis diagnostics. Immunoblots are robust and efficient test systems for rapid simultaneous analysis of multiple antibody specificities. ANCA-3-Line combines these advantages of immunoblot technology with selected, purified antigens.
PR3 is the most frequent component of cANCA and the landmark autoantigen in granulomatosis with polyangiitis (GPA, formerly named Wegener’s granulomatosis) with a clinical specificity of more than 95% for the disease. MPO, the main target antigen of pANCA, is present in 70% of patients with microscopic polyangiitis (MPA) and differentiates MPA from other autoimmune diseases. Autoantibodies against glomerular basement membrane (GBM) indicate Goodpasture syndrome.
Verification of the blot results and subsequent quantitative determination of these three individual ANCA can then be carried out with our specific individual ELISA tests: Anti PR3, highly sensitive Anti-PR3-hs, Anti-MPO, and Anti-GBM.
