Anti-DGP IgG

The Anti-DGP assays by ORGENTEC are ELISA test systems for the quantitative detection of antibodies against deamidated gliadin protein epitopes (DGP). These assays support the formation of a diagnosis when celiac disease is suspected.

After resorption, gliadin is deamidated by tissue transglutaminase (tTG) in the intestinal mucous membrane. Gliadin fragments (deamidated gliadin peptides) bind to the HLA molecules of antigen-presenting cells, where they are recognized by corresponding T-helper cells in the context of specific HLA molecules. The answer is a widespread immune response that causes antibodies against DGP to be formed in addition to those against tTG.

In sera of CD patients both IgA and IgG antibodies against deamidated gliadin protein epitopes are detected. As a significant number of patients are IgA-deficient, sensitive screening for risk groups includes testing for both IgA and IgG antibodies. By IgA antibodies quantification disease activity can be estimated, and the maintenance of a gluten-free diet can be monitored. Performance of the Anti-DGP Screen recognizing both IgA-deficient and IgA-sufficient celiac patients could reduce test costs by eliminating the need for IgA screening.
