Anti-DGP
Anti-DGP IgA is an ELISA test for the quantitative determination of IgA and/or IgG antibodies against deamidated gliadin peptide epitopes (DGP) in human serum or plasma. The assays Anti-DGP IgA, Anti-DGP IgG, and Anti-DGP Screen are used to support a diagnosis of celiac disease.
After resorption, gliadin from food is deamidated by tissue transglutaminase (tTG) in the gastric mucosa. Gliadin fragments (deamidated gliadin peptides) bind to the HLA molecules of antigen-presenting cells. Once placed in context with specific HLA molecules, the fragments are recognized by corresponding T-helper cells. This results in an extensive immune response that includes formation of antibodies against tTG and DGP.
In serum samples from celiac patients, IgA and IgG antibodies against deamidated gliadin peptide epitopes are found. The IgA antibodies make it possible to determine the level of disease activity and to monitor adherence to a gluten-free diet. Because a substantial percentage of celiac patients are deficient in IgA antibodies, sensitive screening of individuals at risk always also includes testing for IgG antibodies. Anti-DGP Screen detects antibodies of both subclasses, IgA and IgG in a single test step, and thus also identifies celiac disease patients with IgA deficiency.
