Anti-MPO (pANCA)
Anti-MPO is an ELISA test for the quantitative determination of IgG antibodies against myeloperoxidase (MPO) in human serum or plasma.
The anti-MPO assay is an important element in the diagnosis of microscopic polyangiitis (MPA).
MPO antibodies are also found in patients with eosinophilic granulomatosis with polyangiitis (EGPA, Churg-Strauss syndrome) and polyarteritis nodosa. They can be used to serologically differentiate these diseases from granulomatosis with polyangiitis (GPA, formerly Wegener’s disease).
Antineutrophil cytoplasmic antibodies (ANCA) are a group of autoantibodies directed against the cytoplasmic components of the neutrophils and monocytes. Current recommendations for ANCA detection include initial testing by indirect immunofluorescence (IF), followed by an antigen-specific ELISA such as Anti-MPO.
MPO is a lysosomal enzyme of neutrophil granulocytes and monocytes. Antibodies against MPO exclusively recognize native conformational epitopes. The wells of the Anti-MPO ELISA are coated with highly purified human myeloperoxidase that fully retains its native conformation.
