Anti-Neutrophil Cytoplasmic Antibodies (ANCA-Formalin)

Indirect immunofluorescence test for the qualitative determination of anti-neutrophil cytoplasmic IgG antibodies (ANCA) in serum or plasma on formalin-fixed human neutrophil granulocytes.

Indirect immunofluorescence tests for the detection of autoantibodies against cytoplasmic antigens of neutrophil granulocytes (ANCA) facilitate the serological diagnosis of autoimmune small-vessel vasculitis. Our formalin-fixed slides should be used subsequent to a pANCA-positive test result with ethanol-fixed granulocytes. They allow for the differentiation of a true pANCA pattern from antinuclear antibodies (ANA), which result in a similar perinuclear fluorescence pattern on ethanol-fixed granulocytes (ORG 891).
If a granular fluorescence pattern is detected in the cytosol with our formalin-fixed slides, antibodies against myeloperoxidase are present. These may be found in cases of eosinophilic granulomatosis with polyangiitis (EGPA; formerly Churg-Strauss syndrome), microscopic polyangiitis (MPA), or Goodpasture syndrome. Perinuclear fluorescence in formalin-fixed granulocytes is indicative of ANA.
In accordance with the “ANCA Consensus Statement” we recommend further serological diagnostic analysis of the cytosolic (cANCA) or perinuclear (pANCA) immunofluorescence pattern with an antigen-specific ELISA test for the detection of anti-PR3 (ORG 618, ORG 318) and anti-MPO (ORG 519, ORG 219).
